2 Days In Tuscany - August 26 - 28

My wife and I are spending two nights in Cinque Terre and then planning on travelling through Tusany for 3 day and 2 nights on our way to Rome. We are currently thinking of renting a car in La Spezia or Pisa and driving through the area but were unsure of a good itenarary. we would ideally like to stay at 1 B&B and visit multiple towns and wineries. Does anyone have any suggestions of romantic places to stay as well as wineries and towns we should visit.
